    The purpose of this article is to analyse the development of tools to support collaborative learning environments. In our view more effort is needed to examine how the use of scaffolding, improved evaluation systems and following up on the work of students can improve the learning process. We analyse results of research in this area and describe related studies initiated at the Universitat Oberta de Catalunya.El objetivo fundamental de este artículo es analizar el desarrollo de herramientas que permitan mejorar el trabajo colaborativo en entornos virtuales de aprendizaje. Se parte de la hipótesis de que hay la necesidad de crear sistemas que favorezcan el proceso de aprendizaje a través de herramientas de andamiaje y mejorar los sistemas de evaluación y seguimiento del trabajo de los estudiantes. Se analizan los resultados de las investigaciones en este ámbito, algunas propuestas y las líneas de trabajo iniciadas en la Universitat Oberta de Catalunya

    Selection of New Probiotics for Endometrial Health

    Microbiota is a crucial player in gynecologic health, in which bacteria can shift to a dysbiotic state triggering a pathogenic process. Based on an ecological understanding of the problem, the aim of this study is to select a potential probiotic strain to improve female reproductive tract based on its capacity to initially lower pH and to promote the reduction of pathogenic bacteria. Based on this rationale, strain Lactobacillus rhamnosus BPL005 was initially selected for its capacity to reduce in vitro pH levels and produce organic acids. Subsequently, strain L. rhamnosus BPL005 (CECT 8800) was demonstrated to have a protective role on endometrial infections in an in vitro model of bacterial colonization of primary endometrial epithelial cells with Atopobium vaginae, Gardnerella vaginalis, Propionibacterium acnes, and Streptococcus agalactiae. In this model, BPL005 when co-cultured with those pathogens was shown to lower pH and to produce organic acids, being lactic acid the most relevant. The co-cultivation of strain L. rhamnosus BPL005 with tested reference pathogens produced a significant reduction in P. acnes and St. agalactiae levels and a non-significant reduction in A. vaginae and G. vaginalis. The colonization of L. rhamnosus BPL005 in the culture decreased IL-6, IL-8, and MCP-1, heightened in the presence of pathogens, and increased IL-1RA and IL-1 beta. Finally, safety was evaluated showing no signs of cytotoxicity, irritation in vaginal tests, or allergic contact dermatitis potential through the Local Lymph Node Assay. Overall, these results show the potential of L. rhamnosus BPL005 strain as a probiotic in gynecological health

    El procés de convergència vers l"Espai Europeu d"Educació Superior (EEES), iniciat a partir de la declaració de Bolonya, col·loca l"assignatura del Pràcticum en una situació fonamental dins dels plans d"estudi de les titulacions. Això ens obliga a repensar-lo a partir dels nous paràmetres didàctics i organitzatius de l"EEES. El projecte de recerca Millora i reorientació del Pràcticum de Pedagogia vers l"EEES s"ha proposat una revisió del Pràcticum de Pedagogia de la UB des de diferents angles: l"estudi de l"ocupabilitat i la inserció laboral dels titulats de Pedagogia, la gestió i l"organització virtual de les tasques relacionades amb la coordinació del Pràcticum, així com l"elaboració d"un pla docent i un dossier de recursos per als tutors i tutores. En aquest article ens centrem exclusivament en el treball realitzat en relació a l"anàlisi de les competències que el Pràcticum de Pedagogia ha de permetre desenvolupar entre els estudiants.     [Background]: Mastocytosis are rare diseases characterized by an accumulation of clonal mast cells (MCs) in one or multiple organs or tissues. Patients with systemic mastocytosis (SM), whose MCs frequently arbor the activating D816V KIT mutation, may have indolent to aggressive diseases, and they may experience MC mediator related symptoms. Indolent SM with recurrent anaphylaxis or vascular collapse in the absence of skin lesions, ISMs(-), is a specific subtype indolent SM (ISM), and this clonal MC activation disorder represents a significant fraction of all MC activation syndromes. The V560G KIT mutation is extremely rare in patients with SM and its biological and prognostic impact remains unknown. [Case presentation]: A 15-year old boy was referred to our hospital because of repeated episodes of flushing, hypotension and syncope since the age of 3-years, preceded by skin lesions compatible with mastocytosis on histopathology that had disappeared in the late-early childhood. Diagnosis of ISM, more precisely the ISMs(-) variant, was confirmed based on the clinical manifestations together with increased baseline serum tryptase levels and the presence of morphologically atypical, mature appearing (CD117+high, FcεRI+) phenotypically aberrant (CD2+, CD25+) MCs, expressing activation-associated markers (CD63, CD69), in the bone marrow. Molecular genetic studies revealed the presence of the KIT V560G mutation in bone marrow MCs, but not in other bone marrow cells, whereas the screening for mutations in codon 816 of KIT was negative. The patient was treated with oral disodium cromoglycate and the disease had a favorable outcome after an eleven-year follow-up period, during which progressively lower serum tryptase levels together with the fully disappearance of all clinical manifestations was observed. [Conclusions]: To the best of our knowledge this first report of a patient with ISM, whose bone marrow MCs carry the KIT V560G activating mutation, manifesting as recurrent spontaneous episodes of flushing and vascular collapse in the absence of skin lesions at the time of diagnosis, in whom disodium cromoglycate had led to long term clinical remission.Peer Reviewe

    Inborn errors of OAS–RNase L in SARS-CoV-2–related multisystem inflammatory syndrome in children

    International audienceMultisystem inflammatory syndrome in children (MIS-C) is a rare and severe condition that follows benign COVID-19. We report autosomal recessive deficiencies of OAS1 , OAS2 , or RNASEL in five unrelated children with MIS-C. The cytosolic double-stranded RNA (dsRNA)–sensing OAS1 and OAS2 generate 2′-5′-linked oligoadenylates (2-5A) that activate the single-stranded RNA–degrading ribonuclease L (RNase L). Monocytic cell lines and primary myeloid cells with OAS1, OAS2, or RNase L deficiencies produce excessive amounts of inflammatory cytokines upon dsRNA or severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) stimulation. Exogenous 2-5A suppresses cytokine production in OAS1-deficient but not RNase L–deficient cells. Cytokine production in RNase L–deficient cells is impaired by MDA5 or RIG-I deficiency and abolished by mitochondrial antiviral-signaling protein (MAVS) deficiency. Recessive OAS–RNase L deficiencies in these patients unleash the production of SARS-CoV-2–triggered, MAVS-mediated inflammatory cytokines by mononuclear phagocytes, thereby underlying MIS-C
